该细胞来源于用SV40大T抗原转化的人胚胎肾细胞293F（F是Fast-growth的首字母），是一种快速生长、高度可转染的克隆分离株。它是制备高滴度慢病毒的良好宿主，能制造高滴度的慢病毒，能够稳定表达pCMVSPORT6TAg.neo 质粒中的SV40大T抗原，且大T抗原的表达受到人类巨细胞病毒（CMV）启动子的控制，表达水平较高。此外，该细胞还可稳定表达新霉素抗性基因，且新霉素抗性基因的表达受到SV40增强子/启动子的控制，因此可用含有0.5 mg/ml G418的培养基中维持培养。 细胞质控检测（可提供检测报告）： 1) 支原体检测结果：阴性 2) 细菌检测结果：阴性 3) 真菌检测结果：阴性 4) 细胞SV40大T抗原检测：SV40大T抗原阳性 5) STR鉴定结果： Amelogenin: X, X D5S818: 8, 9 D13S317: 12, 14 D7S820: 11, 11 D16S539: 9, 13 vWA: 16, 19 TH01: 7, 9.3 TPOX: 11, 11 CSF1PO: 7, 12 D19S433: 15, 18D21S11: 30.2, 30.2 D18S51: 17, 18 D6S1043: 11, 11 D3S1358: 15, 17 Penta D: 9, 9 D2S441: 11, 15 D8S1179: 12, 14 Penta E: 7, 15 D12S391: 19, 21 D2S1338: 19, 19 FGA: 23, 23 |

人胚肾细胞293FT [HEK293-FT]完全培养液（SCSP-688）配方： DMEM培养基（Gibco, 货号11995065）： 88% 胎牛血清FBS（Gibco）： 10% 非必需氨基酸溶液NEAA (Gibco，货号11140050)： 1% GlutaMAX (Gibco，货号35050061)： 1% 添加G418（Gibco，货号10131027）至终浓度0.5 mg/ml 参考传代比例：1:3 参考传代周期：2-4天 参考换液频率：每2-3天换1次液 冻存液配方：完全培养液90%，DMSO 10% 培养条件：气相：空气95%，二氧化碳5%；温度：37摄氏度。 细胞运输方式：活细胞保温运输或冻存管干冰运输。 注意事项： 1. DMEM培养基中已含有4 mM L-glutamine, 但由于L-glutamine会随着时间的推移而缓慢降解，所以我们额外添加 2 mM GlutaMAX。 2. 细胞复苏时可选用不含G418的完全培养基，第二天更换为含有0.5 mg/ml G418 的新鲜完全培养基。 |
