据报道，4T1是一种三阴性乳腺癌细胞系（ER−,PR−, HER2−）。它是一株源自未经诱变剂处理的410.4肿瘤中筛得的6-硫鸟嘌呤抗性细胞系。4T1细胞在BALB/C小鼠体内的肿瘤生长和转移扩散与人乳腺癌非常相似。这种肿瘤是人类乳腺癌第四阶段的动物模型。当注射到BALB/C小鼠体内时，4T1会自发产生高转移性肿瘤，在原发肿瘤原位生长的同时转移到肺、肝、淋巴结和大脑。4T1诱导的肿瘤既可以作为术后模型，也可以作为非手术模型，因为4T1诱导的肿瘤在两个具有相似动力学的模型中自发转移。我库冻存时为P22代（该代数为细胞建系以来的准确代数）。 细胞质控检测（可提供检测报告）： 1) 支原体检测结果：阴性 2) 细菌检测结果：阴性 3) 真菌检测结果：阴性 4) STR鉴定结果无误，结果为： ①该株细胞DNA进行小鼠细胞STR分型结果显示，扩增后图谱清晰，分型结果良好，与数据库4T1匹配率为100%：1-1：15, 16；1-2：17；2-1：16, 17；3-2：14, 15；4-2：21.3；5-5：14；6-4：18；6-7：12；7-1：25.2；8-1：13；11-2：18, 20；12-1：16；13-1：16.2；15-3：22.3；17-2：15；18-3：18, 19；19-2：13；X-1：25。 ②该株细胞确为小鼠细胞，没有人源细胞污染。 |

| 培养基和添加剂 Complete Growth Medium and Culture Conditions |
小鼠乳腺癌细胞4T1完全培养液（目录号：SCSP-653）配方： RPMI 1640培养基 (Gibco, 货号11875093)：90% 胎牛血清FBS (Gibco)：10% 参考传代比例：1:6-1:8 参考传代周期：2-4天 参考换液频率：每周2次 冻存液配方：完全培养液95%，DMSO 5% 培养条件：气相：空气95%，二氧化碳5%；温度：37摄氏度。 细胞运输方式：活细胞保温运输或冻存管干冰运输。 注意事项：该细胞培养中不要使其生长至完全融合，应该在达到90%融合度时传代。吸弃培养基，用0.25%胰蛋白酶（含0.53 mM EDTA）溶液冲洗。吸走溶液，再添加1到2毫升胰蛋白酶EDTA溶液。让培养瓶在室温（或37°C）下放置，直到大多数细胞从培养瓶底面脱落。加入新鲜培养基，吹打均匀并分配到新的培养瓶中。 |
