人肝癌细胞Hep G2 [HEPG2]是1975年从一名罹患肝癌的15岁白人男性青年的肿瘤组织中分离获得。据报道,该细胞不携带乙肝病毒基因组,但表达一系列蛋白,包括甲胎蛋白、白蛋白、α-2-巨球蛋白、α-1-抗胰蛋白酶、转铁蛋白、α-1-抗凝乳蛋白酶、结合珠蛋白、铜蓝蛋白、纤溶酶原、补体C4、C3激活物、纤维蛋白原、α-1酸性糖蛋白、α-2-HS-糖蛋白、β-脂蛋白、视黄醇结合蛋白等;表达胰岛素受体和胰岛素样生长因子IGFⅡ的受体,表达3-羟基-3-甲基戊二酰还原酶(3-hydroxy-3-methylglutaryl-CoA)和肝三酰甘油脂肪酶活性。另据资料显示,该细胞在免疫抑制小鼠中不致瘤,在半固体培养基中可成瘤,可用于大规模培养体系,已广泛应用于细胞毒理学实验和肿瘤生物学研究。 细胞质控检测(可提供检测报告): 1) 支原体检测结果:阴性 2) 细菌检测结果:阴性 3) 真菌检测结果:阴性 4) 经STR检测结果无误。STR结果如下:D5S818: 11,12;D13S317: 9,13;D7S820: 10,10;D16S539: 12,12;vWA: 17,17;THO1: 9,9;Amelogenin: X,Y;TPOX: 8,9;CSF1PO: 10,11 |

人肝癌细胞 Hep G2完全培养液(目录号:SCSP-651)配方: MEM培养基 (Gibco,货号11095080):89% 胎牛血清FBS (Gibco):10% 非必需氨基酸溶液NEAA (Gibco,货号11140050):1% 参考传代比例:1:3 参考传代周期:3-5天 参考换液频率:2-3天换液一次 冻存液:完全培养液95%,DMSO 5% 培养条件:气相:空气95%,二氧化碳5%;温度:37摄氏度。 细胞运输方式:活细胞保温运输或冻存管干冰运输。 【注意事项】: 1. Hep G2 [HEPG2]对培养条件要求较严格,特别是 pH 值和血清质量,否则可能影响细胞凝集数量,应使用高质量低内毒素,未灭火的胎牛血清,可以帮助圆形的细胞丛簇更好的贴壁及形成单层细胞。细胞内容易有空泡,特别是在融合时,这属于正常现象。 2. 该细胞部分团块生长,复苏后可能存在形态不典型的情况,经几次传代后,上皮细胞形态会更加典型。 3. 该细胞对血清质量较为敏感,我库建议您使用优质胎牛血清进行培养或选择订购我库配套MEM完全培养液(目录号为SCSP-651)。 |
