NIH/3T3是从小鼠NIH/Swiss品系的胚胎中分离并建立的成纤维细胞株。与其它3T3细胞的建系方法一样,NIH/3T3细胞是从NIH Swiss小鼠胚胎培养物中建立的高度接触抑制的连续传代细胞株。为了培育在形态学特征上更适合于进行转化分析的亚株,建立的NIH/3T3细胞株又进行了5轮以上亚克隆。该细胞易于转染,对DNA转化及转染研非常有用。据报道,该细胞对肉瘤病毒的转化灶形成和白血病病毒增殖高度敏感,鼠痘病毒(ectromelia virus,ECTV)检测阴性。 细胞质控检测(可提供检测报告): 1) 支原体检测结果:阴性 2) 细菌检测结果:阴性 3) 真菌检测结果:阴性 4) STR鉴定结果: ①该株细胞DNA进行小鼠细胞STR分型结果显示,扩增后图谱清晰,分型结果良好: 1-1: 10;1-2: 13, 17;2-1: 9;3-2: 14, 15;4-2: 19.3, 20.3;5-5: 14, 15;6-4: 15.3;6-7: 12;7-1: 29;8-1: 15;11-2: 15, 17;12-1: 20;13-1: 16.2;15-3: 20.3;17-2: 13, 14;18-3: 17, 19;19-2: 11, 12;X-1: 25。 ②该细胞与ExPASy中NIH 3T3细胞的STR数据匹配率为100%。 ③该株细胞确为小鼠细胞,没有人源细胞污染。 |

小鼠胚胎成纤维细胞NIH/3T3 完全培养液(目录号:SCSP-646)配方: DMEM培养基 (Gibco, 货号11995065):90% bovine calf serum,新生牛血清 (四季青,22012-8612;或Gibco,26010074):10% 参考传代比例:1:3-1:4 参考传代周期:2~3天 参考换液频率:2~3天,传代时换液 冻存液配方:完全培养液95%,DMSO 5% 培养条件:气相:空气95%,二氧化碳5%;温度:37摄氏度。 细胞运输方式:活细胞保温运输或冻存管干冰运输。 【注意事项:】 1. 不可使NIH/3T3细胞生长过密至完全汇合(细胞将失去接触抑制的特性)。细胞密度达到80%即可传代,每周至少传代2次。 2. 该细胞对于血清较为敏感,不使用胎牛血清(FBS)培养该细胞。 |
