Description
HT-29是一种具有上皮形态的细胞系,于1964年从一名44岁白人女性结直肠癌患者的原发肿瘤中分离出来。该细胞系是一种适宜的转染宿主,在肿瘤和毒理学研究中具有应用价值。 用移植培养方法和含15%FBS的F12培养液从原发性肿瘤分离而来。近年来,已建株的培养细胞用含相同血清的McCOY's 5A培养液进行培养。该细胞系在裸鼠中致瘤,也能在类固醇处理的地鼠中致瘤。该细胞可合成IgA、CEA、TGFβ结合蛋白和黏液素,同时呈现出癌基因c-myc、K-ras、H-ras、N-ras、Myb、sis、fos阳性;p53基因过表达,并且在p53基因的第273位密码子处发生G突变。细胞引进入库精准代数为P128代。

McCOY's 5A (Gibco货号:16600082),90%;优质胎牛血清(GIBCO),10%。

气相:空气,95%;二氧化碳,5%。
温度:37摄氏度。

培养参考:细胞消化时间8-10min(0.25%胰酶)。传代比例1:2,传代周期2-3天
冻存液配方:完全培养液92%,DMSO 8%

细胞运输方式:活细胞可保温运输或干冰冻存管。

注意事项:
HT-29细胞呈块片状聚集生长,细胞胞体黑点较多是正常现象。
